STRUCTURAL BIOLOGY AND BIOPHYSICS SECTION
The Structural Biology and Biophysics Section offers access and services for technologies in structural studies, molecular interactions and biophysical characterizations of a wide range of biomolecules.
The protein purification facility is equipped with FPLCs and a wide assortment of chromatography columns, covering the conventional protein purification techniques including affinity, ion exchange and size-exclusion chromatography. The purification process includes application to column, elution, buffer exchange (if necessary), desalting, sterile filtration and OD measurement.
The structural biology facility covers protein crystallization and structure determination by X-ray crystallography and nuclear magnetic resonance (NMR) spectroscopy as the main methods for high resolution structure determination. We provide a complete package for protein structure determination, from protein purification, crystal screenings to X-ray diffraction analysis.
The high-field NMR facility offers multinuclear NMR service for a range of biomolecules. Typical experiments include COSY, NOESY, TOCSY, HSQC & HMBC. Other than structure determination in solution, the NMR facility offers services in protein-ligand interaction studies as well as metabolomics analysis.
The biophysics facility provides a platform for molecular interactions and characterization study. Nano ITC provides thorough information of molecular interactions through thermodynamic approach in the form of binding constant, stoichiometry and enthalpy. Biacore X 100 provides binding constant information for immobilized macromolecules, particularly for drug screening and development of biosensor. The biophysics facility is also equipped with a few spectroscopy instruments namely circular dichroism, fluorescence spectrometer and UV/Vis spectrometer.Services
